Modular Polyketide synthases (PKSs) are a family of multi-emzyme complexes tht synthesize the polyketide cores of a multitute of biologically imporatant pharmaceuticals. We have performed an extensive analysis of characterized modular PKSs and found insight into their mechanism of substrate specificty and stereo-selectivity. We have developed a computational tool that can help identify and analyse PKS domains in various, hitherto uncharacterized clusters. It can also assist in rational genetic engineering of novel antibiotics.
